阿片受體家族包括κ-阿片受體（KOP）、 δ-阿片受體（DOP）、µ-阿片受體（MOP） 和痛覺肽阿片受體（NOP），均屬于 G 蛋白偶聯(lián)受體，主要通過偶聯(lián)下游 Gi 蛋白發(fā)揮功 能。阿片受體在疼痛感知中起作用，并介導(dǎo)合成阿片的低運(yùn)動(dòng)、鎮(zhèn)痛和厭惡作用。這種基 因的變異也與酒精依賴和成癮有關(guān)。 | |
II. Description | |
OPRK1 β-Arrestin CHO OPRK1 β-Arrestin CHO 報(bào)告基因藥靶模型很好的模擬了體內(nèi) OPRK1/β-Arrestin 的信號(hào) 轉(zhuǎn)導(dǎo)過程，原理見下圖所示。當(dāng)缺乏配體刺激時(shí)，β-Arrestin 不與 OPRK1 結(jié)合，融合β- Arrestin 的熒光素酶處于失活構(gòu)象，當(dāng) OPRK1 遭遇配體刺激時(shí)，融合熒光素酶報(bào)告基因的 β-Arrestin 被招募，使熒光素酶報(bào)告基因處于激活狀態(tài)，加入其底物后發(fā)光信號(hào)增強(qiáng)。 | |
